生物活性:CD137(也称作 4-1BB),CD137 是一种可诱导的细胞表面受体,属于肿瘤坏死因子受体( tumornecrosis factor receptor, TNFR)超家族成员,由肿瘤坏死因子受体超家族成员 9(TNFRSF9)基因编码。广泛表达于激活后的 T 细胞、NK 细胞等人体免疫细胞的表面。它的配体是 CD137L,也属于 TNFSF 成员,是在活化的 CD4+ 和 CD8+ T 细胞、NKT、NK 细胞、DC、巨噬细胞、嗜酸性粒细胞、嗜中性粒细胞和肥大细胞表达的诱导型共刺激受体,特别是在 CD8+ T 细胞上。CD137/CD137L 通过在免疫细胞间传递活化、增殖或者凋亡信号来调节 T 细胞介导的免疫反应。CD137 的活化信号转导可以刺激 NK 细胞和 T 细胞增殖并产生抗肿瘤活性,并产生持久的记忆应答。利用激活型 4-1BB 单抗激活 4-1BB,4-1BB 募集 TNFR 相关因子 TRAF1 和 TRAF2 形成异源三聚体,通过 c-Jun N 末端激酶(JNK)途径,细胞外信号调节激酶(ERK)途径等加强信号转导,并通过主转录因子 NF-κB 调节 4-1BB信号,刺激 T 细胞和抗原提呈细胞增殖并分泌细胞因子,提高机体的抗肿瘤免疫应答水平。
