程序性细胞死亡蛋白1 (PD-1)也被称为CD279和PDCD1，是I型膜蛋白，是扩展的T细胞调控因子CD28/CTLA-4家族的成员。PDCD1在活化的T细胞、B细胞、巨噬细胞、髓样细胞和一部分胸腺细胞的表面表达。PD-1有两个配体PD-L1和PD-L2，都是B7家族的成员。几乎所有的小鼠肿瘤细胞系，包括PA1骨髓瘤，P815乳腺细胞瘤和B16黑色素瘤在IFN-& γ;治疗后表达PD-L1。PD-L2表达受限，主要在dc和少数肿瘤系中表达。PD1抑制t细胞增殖和相关细胞因子的产生，包括IL-1、IL-4、IL-10和IFN-& γ;通过抑制PI3K/AKT通路的激活和转导。此外，PD1的结合通过去磷酸化关键信号转换器抑制bcr介导的信号。在体外，PD-L1-Ig治疗抗cd3刺激的T细胞可减少T细胞增殖和IFN-& γ;分泌。针对增强免疫系统的PD-1的单克隆抗体正在开发，用于癌症研究。
