生物活性：ED50 < 0.5µg/ml，用NIH-3T3细胞在小鼠Klotho 1.25µg/ml和肝素10µg/ml存在下进行细胞增殖试验，对应的比活性为> 2.0 × 103单位/mg。与FGF-19/15和FGF-23一起，FGF-21被归类为非典型FGF亚家族的成员，因为它必须与Klotho共受体复合，才能与FGF受体结合并激活下游信号通路。在体内，FGF-21在肝脏、胰腺、脂肪组织和骨骼肌中表达，并在能量代谢中起核心作用。FGF-21的表达受游离脂肪酸和胰岛素抵抗状态的刺激，并与全身胰岛素抵抗相关。FGF-21激活脂肪细胞的葡萄糖摄取并增加胰岛素敏感性，这意味着它是一个具有潜在抗糖尿病特性的新靶点。
