重组人RANKL是肿瘤坏死因子(TNF)超家族的II型膜蛋白,影响免疫系统,控制骨再生和重塑。RANKL为核因子(NF)-κB (RANK)配体。当RANKL与RANK结合时,会发生三聚体化,然后与接头分子TNF受体相关因子6 (TRAF6)结合。这导致多个下游信号级联的激活,包括nf - κ b、丝裂原活化蛋白激酶(MAPK)、活化蛋白1 (AP-1)和活化T细胞核因子(NFATc1),导致多核骨吸收破骨细胞的形成。RANKL广泛表达于骨骼肌、胸腺、肝脏、结肠、小肠、肾上腺、成骨细胞、乳腺上皮细胞、前列腺、胰腺等。
2μg/ml (100 μl/孔)固定化RANKL可与Human OPG-His结合。
RANKL的ED50为0.15 ug/ml。
